The current one just brings it to the next challenge, but if you’re doing player puzzles in the order of length, or by # of players solved, this gets annoying. So, I think the system should be the next one in the list of what you have the puzzles sorted to. [3:05 PM]

steven123505: There’s a problem when a link is posted in chat, it will replace a tab of your browser. For example, if you have something like youtube open in another tab, sometimes, when you click on the link, the link would replace the youtube tab. [3:06 PM]

Edward_Lane: oh can someone explain what th e'glowing bits' represent in terms of actual aptamer binding sites - if the predicted structure with teh aptamper does not have all the glowing bits around one region does that mean that the aptamer doesn't really have a good binding site ? [3:14 PM]

Brourd: when the binding site is not "bound" it will glow and pulse red, which is an indication that the energy bonus is not applied to the design [3:17 PM]

Brourd: The predicted results just pair/ do not pair based on the reactivity scores [3:19 PM]

Edward_Lane: yes but some bases are still marked with the glow [3:19 PM]

Brourd: it can throw off the prediction [3:19 PM]

Edward_Lane: and sometimes the marked bases are not in one spot [3:20 PM]

Brourd: but of course, all bases that are a part of the aptamer glow [3:20 PM]

Brourd: you are probably just using the estimated prediction of how the RNA folds a little bit too much. Since an RNA switch, by definition, has multiple conformations it can fold into, the SHAPE data will end up a bit "murky" [3:21 PM]
